From here, the actual sequencing can begin. Primers and fluorescently-labeled chain terminators are added to the reaction mixture, resulting in the addition of one nucleotide to each strand of DNA on the lawn. This process proceeds until the reaction is complete, resulting in massively parallelized sequencing. The short reads acquired through this process can be combined via a computational technique called paired end analysis, which links reads by analyzing overlapping sections, to generate the whole sequence.
